By analyzing gene regulation in endothelial cells, the researchers found that RNF20 controls RNA polymerase II pausing and mRNA processing, particularly affecting VEGFA signaling and Notch-dependent gene expression. These processes are crucial for balanced vessel growth, as RNF20 loss leads to abnormal tip cell specification. The findings highlight RNF20’s essential role in coordinating VEGF–Notch signaling circuits, providing insights into potential therapeutic strategies for angiogenesis-related diseases.
